Unlike some other games RDR2 only has one autosave slot by default, which is why that pop-up can get a little excessive if you reboot the game or (re)load saves often as it automatically asks for permission everytime.Autosave does not overwrite a manual save. When the game asks if you want autosave to save file, it is because a manual save was loaded, and it's asking about overwriting the existing autosave file.
